Choosing the correct syringe filter for HPLC/UHPLC sample filtration can sometimes be challenging. There are many different membrane choices and devices in the marketplace. Pall have simplified syringe filter selection by offing Acrodisc® syringe filters with universal membranes that are suitable for all HPLC/UHPLC analytical sample preparation filtration requirements.

Sample filtration is a simple and economical practice that serves to extend the life of consumable HPLC parts, decrease system wear and tear, and preserve the integrity of the HPLC system. Filtering a sample prior to sample injection into a HPLC will help protect sensitive parts of the instrument from particulate build up and reduce system downtime. Sample filtration using syringe filters containing 0.45 μm (HPLC) or 0.2 μm (UHPLC) pore size membranes will also extend the life of chromatography columns.
High performance columns are composed of small particles of narrow size distribution. Optimal peak profiles depend on column operating characteristics and should be instrument independent. Columns, depending on sample type, sample preparation, and operator filtration practices, can handle a few to several thousand injections. Two significant problems with HPLC columns are chemical and physical changes. Chemical changes are prevented with guard columns. Physical changes involve blocked frits and channel voids created by particulate matter build up.
Pall’s Acrodisc syringe filters with universal membranes have been designed to protect analytical instrumentation and improve the reliability of results. These syringe filters contain the universally solvent compatible wwPTFE (water wettable Polytetrafluoroethylene) membrane and are suitable for use in both aqueous and organic applications.
These syringe filters are constructed with solvent compatible polypropylene material. The polypropylene plastic provides the syringe filter with exceptional strength, it is the highest grade of plastic with minimal additives, and passes United States Pharmacopeia (USP) Biological Reactivity Test, In Vivo <88> plastics testing.
Both the wwPTFE membrane and polypropylene housing are extremely low in levels of potential extractables that can lead to chromatographic interference. The devices are low in protein and API binding which make them ideal for drug studies, such as dissolution testing.
Pall provides HPLC certification with these products, ensuring that analytical results will not be compromised by extractable filter materials. Pall membranes are tested for compatibility with common HPLC solvents including water, methanol, and acetonitrile, using established HPLC procedures. In addition, to verify low levels of UV-detectable extractables, samples of the syringe filters are evaluated prior to release.
Pall’s Acrodisc syringe filters with universal membranes are available in 13 mm and 25 mm diameters to suit the volume requirements of different applications. The 13 mm devices feature Pall’s proprietary minispike outlet configuration, it offers a lower hold-up volume and easier filtration into autosampler vials compared to standard luer outlets.
The 25 mm device, Acrodisc One™ syringe filter with wwPTFE membrane, sets the standard for HPLC/UHPLC analytical filtration. It can extend the life of a HPLC column by up to 52 times over unfiltered samples.
Effects of filters on HPLC column life following injections of unfiltered and filtered 0.05% latex sphere suspensions. With unfiltered samples, the column failed due to plugging after 19 injections. Samples passed through competitor filters plugged the columns after 500 injections. No increase in backpressure was observed after 1000 injections of samples filtered with Pall Acrodisc One™ syringe filters.
Acrodisc One syringe filters are available with either 0.45 μm (HPLC) or 0.2 μm (UHPLC) pore size wwPTFE membranes. For solutions with high particulate load Pall supply a version of the Acrodisc One with a multi-layered glass fiber prefilter (GxF) above the wwPTFE membrane. Syringe filters with the GxF configuration prefilter provide two to four times the throughput of standard prefilter devices for particulate-laden samples.
Acrodisc One syringe filters have been designed and certified to meet the exacting requirements of automation systems. They demonstrate critical criteria including smooth filter-to-filter release, consistent turret advancement, exceptional housing strength and a strict outside filter geometry to ensure proper alignment and consistent operation. AutopackTM tube packaging is available for compatible automated equipment.
